Tumor‐Targeted Poly(ArgGlyAsp) Nanocapsules for Personalized Cancer Therapy – In Vivo Study

The arginine-glycine-glutamic acid (RGD) sequence, an αvβ3 integrin recognition site, is overexpressed in malignancies and neovasculature, making it a potential therapeutic target. Herein, efficacy/safety of tumor-targeted RGD-based proteinoid nanocapsules (NCs) entrapping synergistic combination two drugs—palbociclib (Pal), CDK4/6 inhibitor, alpelisib (Alp), P13K as cancer treatment, assessed. P(RGD) polymers are produced by thermal step-growth polymerization R, G, D under inert atmosphere. NCs, hollow encapsulating 25 w% each Pal Alp, formed self-assembly the polymer. encapsulation yields Alp 72% 95%, respectively. Long-term stability, controlled release, cellular uptake,synergistic cytotoxicity, induced cell death evident from vitro experiments. Findings vivo breast, colon gastric patient-derived xenograft (PDX) mice experiments consistent with studies showing that response to treatment drug-loaded NCs similar elicited free drugs, reduced side effects. study demonstrates clinical utility for treatment.
